MDL-001 is a first-in-class, oral, non-nucleoside, direct-acting broad-spectrum antiviral targeting the RdRp Thumb-1 cryptic site, a highly conserved allosteric choke point in viral RNA-dependent RNA polymerase shared across positive-sense single-stranded RNA viruses. The single molecule demonstrates potent antiviral activity across six viral families: HCV, HBV, HDV, SARS-CoV-2, Influenza A/B, and RSV, including clinically challenging co-infections (HCV/HBV, HBV/HDV). Designed to overcome the dose-limiting thrombocytopenia that hindered prior pan-BET inhibitors. Indications span fibrosis, immunology & inflammation, cardiovascular, and oncology. IND targeting 2027.
